Xavier University is the fourth oldest Jesuit university in the United States and the sixth oldest Catholic university in the United States. The school was founded in 1831 as a man's dance street next to St. Francis Church in downtown Cincinnati. On October 17, 1831, the church, then known as the Temple of Athena, was patronized by Bishop Edward Fenwick to Saint Francis Xavier. At the request of Bishop John Baptist Purcell, the Jesuits took over the Temple of Athena (The Athenaeum) in 1840 and changed its name to the College of Saint Xavier to commemorate Spain in the 16th century Saint Francis Xavier, the Jesuit missionary, is a Spanish Basque like Ignatius Loyola, the founder of the Jesuit.

Saint Xavier College purchased 26 acres (0.11 km2) of land from the Avondale Sports Club in 1912 and moved to Norwood, which is currently located about 5 miles (8.0 km) north of downtown Cincinnati. The "primitive" Anthenaeum is now the Archdiocese of Cincinnati Theological Seminary. Saint Xavier College and Saint Xavier High School were formally separated in 1919, but it was not until 1934 that they achieved financial independence. In 1930, the name of the school was changed to the current name Xavier University for the second time.

The Williams School of Business was established in 1961, and Xavier's first PhD program in psychology began in 1997. Xavier fully admitted women in 1969, but women began to attend evening, weekend, and summer school in 1914. Edgecliff College is another Catholic college in Cincinnati that merged with Xavier University in 1980.

In 2000, Xavier opened the door to the Cintas Center, an arena for musketeers. Xavier also opened the Gallagher Student Center in 2002.

Smith Hall and Conaton Learning Commons opened in 2010 as part of James E. Hoff, SJ, Academic Quadrangle. Father Hoff was the 33rd president of the university from 1991-2000. Father Michael J. Graham, SJ, Hoff's successor and 34th president, still serves Xavier. Fenwick Place is a residential area that opened in autumn 2011.
